Due to their asymptomatic development as well as the lack of sensitive and specific screening methods, the disease is diagnosed in advanced and incurable stages. Although tumors usually respond to first line taxanes and platinum-based chemotherapy, most patients develop relapse and chemoresistance. Inflammation has been involved in the initiation and development of many types of cancer, including HGS-OC. Recently, the protein components of the innate immune system, known as inflammassome, have been associated with the mechanisms of progression and metastasis of neoplasias, however, it’s role in drugresistance is little explored. In this study, through RT-PCR and western blot analysis we investigated the clinical significance of inflammassomas in modulating the chemoresistant phenotype in cisplatin-resistant cells (ACRP) obtained from the parental line of HGS-OC (A2780). Our results have shown that, even though mRNA expression levels of NLRP3 remained unaltered in the ACRP, the NLRP3 protein expression appears to vary in a dose-dependent way with cisplatin treatment, although it does not configure the main event in chemoresistance. An increase in mRNA expression levels of the NLRP1, but not of NLRC4, in resistant cell line ACRP was also obtained. In conclusion, our study was the first to show the involvement of NLRP1 inflammassome in the development and regulation of cisplatin-resistance in HGS-OC cells, making it an attractive target with great therapeutic potential against the disease.

The most common one, the ischemic stroke, is caused by the presence of thrombus or embolus in the vessel, or because of a cardiac arrest. Physical and cognitive impairment are recurrent sequelae in cerebral ischemia, and can be minimized with the adoption of appropriate post-stroke rehabilitation treatments, such as physical exercise (PE) and enriched environment exposure (EE). In this context, this research investigated the association between EE exposure and PE in motor and cognitive function recovering in mice submitted to an ischemic stroke. The animals used in the experiments, male mice C57Bl/6 (8 to 12 weeks old) were either submitted to bilateral occlusion of the common carotid arteries (BCCAo) for 35 minutes or sham-operated (sham). Subsequently, ischemic and sham animals were splited into one between eight groups, according to, housing conditions (standard environment (SE) or enriched environment (EE)) and whether or not they underwent conditioned physical activity sessions (PE): ischemic or sham treated with SE without PE (SE/S), PE in standard environment (SE/E), association of EE and PE (EE/E), e standard environment without PE (SE/S). Rehabilitation started 48 h after surgery and lasted for 30 days. The animals in the EE groups were placed in large boxes (60cm × 50cm × 22cm) with 6 toys alternated weekly, while the others were placed in standard mice boxes. The PE model used was swimming, performed 3 times a week for a period of 4 weeks. The duration of the sections was gradually increased each week, until a maximum time of 30 minutes. The statistics indicate that the animals submitted to BCCAo developed motor deficits. In addition, the animals that were submitted to the EE and PE association obtained an increase in motor, but it was not possible to conclude that there was an improvement in cognitive function. In this way, this study suggests that the association between EE and PE is more effective for motor sequelae treatment of animals submitted to BCCAo.

Traumatic events during this period may impair physiological and psychological development, potentially causing short- and long-term changes. Maternal deprivation (MD) is a well-established protocol used to investigate both neurobiological and behavioral changes such as anxiety disorders. It has been demonstrated that environmental enrichment (EE) protocols promote numerous sensory, motor, and cognitive benefits in laboratory animals, and may be used to intervene in changes caused by postnatal adverse events and to prevent the occurrence of psychiatric disorders in adulthood. In this context, we evaluated the implications of EE as a strategy to prevent the maternal deprivation effects on anxiety behaviors and gene expression of the serotonergic system components.

Furthermore, stressful experiences stimulate abusive consumption of ethanol, which induces neuronal damages and contributes to increases on parameters of oxidative stress on Central Nervous System. The aim of this research was to evaluate the effects of Maternal Separation (MS) and of intermittent ethanol binge drinking in adolescent rats on behavioral and oxidative stress parameters. Pregnant Wistar rats were used. After birth of litters, the animals were divided into control group (NS) or maternal separation group (MS). Maternal separation was performed from 2 to 15 postnatal day, 3 hours daily. At postnatal day 35, animals were divided again into 3 groups: animals which received intragastric saline (vehicle group) or ethanol in doses of 3.0 or 6.0 g/kg. Ethanol was administered once a day, in a regimen of two consecutive days interspersed by two days without ethanol, totalizing 10 doses for chronic binge treatments. Acute binge exposure happened at postnatal day 35 until 37 postnatal day.

Studies have shown that cyclodextrins can form promising interactions with proteins, which has several applications in biotechnology and biochemistry, such as the solubilization of hydrophobic compounds and the protection of compounds sensitive to thermal, oxidative degradation, among others. In this work, in order to evaluate the influence of this polysaccharide on conformational stability and protein activity, physicochemical tools were used, adopting bovine trypsin as an enzymatic model of study, due to the knowledge of its structure and enzymatic activity. This enzyme belongs to the class of serine proteases and has several isoforms, α-trypsin, β-trypsin and ψ-trypsin being the most studied. The results showed that protease amidasic activity increased up to about 76% in commercial trypsin, 82% in alpha isoform, and 45% in psi isoform, suggesting that the addition of β-cyclodextrin optimized the amidsis activity of the trypsin isoforms tested. Fluorescence and absorption spectroscopy revealed changes in the structural properties of trypsin due to the interaction with βcyclodextrin, affecting the activity and conformational stability of trypsin. The formation of supramolecular states of trypsin isoforms in the presence of β-cyclodextrin by means of dynamic light scattering (DLS) was also evaluated, in which an increase in the size and dispersivity of the molecule suggestive of the formation of supramolecular states was verified. The joint evaluation of these results suggested that there was the formation of stabilizing interactions between β-cyclodextrin and trypsin isoforms through the formation of supramolecular states, with possible formation of inclusion complexes. Thus, it is suggested that β-cyclodextrin may interact with trypsin and affect its proteolytic activity and stability and thus the influence of the osmolyte can be understood at the molecular level.

Furthermore, was analysed antioxidant activity and citotoxicity of AuNPs, for investigate your potential biological application. For this goal, the best capping time, and subsequently the optimum pH range and final extract concentration was determined and was realizes a 32 factorial design with 3 levels and 2 variables (pH and extract concentration) to verify the optimal capping conditions. In this way, it has been proved the adsorption was most intense between pH 3 and 5 and the most stable was the lower final extract concentration. The x-ray diffraction and the transmission electron microscopy confirmed the face-centered cubic structure of citrate AuNPs and the quasi spherical aspect, respectively. The adsorption of aromatic molecules was confirmed by spectroscopy techniques: Fourier-Transform Infrared (FTIR) and Raman. The DPPH and FRAP analyzes shows enhancement of antioxidant activity with capping of citrate gold nanoparticles. Furthermore, the citotoxicity assay shows low citotoxicity of capped gold nanoparticles.

- ItemCaracterização bioquímica e estrutural da proteína catecol o-metiltransferase (COMT) como potencial alvo para drogas contra paracoccidioidomicose(Universidade Federal do Espírito Santo, 2018-03-28) Cruz, Fabiano Torres; Santos, Alexandre Martins Costa; Gonçalves, Juliana Barbosa Coitinho; Campos, Fabiana Vasconcelos; Costa, Mariana Amália FigueiredoParacoccidioidomycosis (PCM), a systemic mycosis caused by fungi of the genus Paracoccidioides, is endemic in Latin America, with Brazil being the most affected country. Currently, treatment with traditional antifungals is usually long, costly, restricted to a few classes of drugs and with cases of resistance already reported. The catechol O-methyltransferase (COMT) protein, which has no structure solved in Paracoccidioides or other fungi, catalyzes the methylation of a catechol substrate using S-adenosylmethionine and Mg2+ ions as cofactors and appears to participate in important processes in pathogenic fungi. Thus, its biochemical and structural characterization could contribute to the development of new and more effective drugs for the treatment of PCM. To study COMT, its coding sequence was cloned into the pET-28aTEV vector, which adds an N-terminal histidine-tag (His-tag) to the expressed protein. The recombinant vector was then inserted into the strains of Escherichia coli BL21, ArcticExpress, Rosetta and Rosetta-gami. Recombinant COMT was expressed in the soluble fraction of ArcticExpress and it was subjected to nickel affinity chromatography followed by gel filtration (Ultrahydrogel and Superose 12 columns). Western blot with anti-Histag antibody showed the two major bands observed on SDS-PAGE (30 and 60 kDa) of the fraction obtained after affinity. The gel filtration profile showed three main peaks (fractions 1, 2 and 3) which, on SDSPAGE, showed the presence of a prominent band near 60 kDa. Samples from the three fractions were used in an activity assay to evaluate the catechol consumption at 35 ºC and pH 7.5, and fraction 2 showed a higher activity/protein ratio. Theoretical calculations of the masses of gel filtration chromatography peaks indicate that COMT behaves as a dimer. Fraction 2, although not pure, was subjected to thermal denaturation test, where a process of aggregation or denaturation was detected at temperatures above 320 K (~47 ° C). Circular dichroism of fraction 2 indicated an αhelix and β-sheet profile different from other COMT. An in silico model has also been generated and the structural data obtained can contribute as a guide for future biochemical characterization.

- ItemCaracterização bioquímica e farmacológica da Sp-CTx- Toxina Multifuncional da Peçonha do Peixe Escorpião Scorpaena Plumieri(Universidade Federal do Espírito Santo, 2017-03-30) Malacarne, Pedro Felipe; Figueiredo, Suely Gomes de; Pires, Rita Gomes Wanderley; Guimarães, Marco Cesar Cunegundes; Trindade, Juliana Bambrilla CarnielliCytolysins are substances elaborated by many different organisms that exhibit toxic activities on cells, causing its dissolution through cytolysis. Fish venom cytolysins are multifunctional proteins that display neurotoxic, cardiotoxic and inflammatory effects, described as "protein lethal factors". A pore-forming labile hemolytic cytolysin called Sp-CTx (Scorpaena plumieri - Cytolytic Toxin) (glycoprotein, 2 subunits of 65 kDa) was recently purified from the venom of the scorpionfish Scorpaena plumieri. The aim of this work was to establish optimal storage conditions to maintain the activities of this toxin and to continue its biochemical and pharmacological characterization. For this, SpV (Scorpaena plumieri Venom) and Sp-CTx were stored in different conditions (pH, temperature and in the presence of co-solutes), for different periods of time. Afterwards, their hemolytic activity on rabbit erythrocytes was compared to the activity of the freshly extracted venom and purified toxin. We also evaluated the effects of different cations and membrane lipids on the hemolytic activity of Sp-CTx. It was demonstrated that storage of Sp-CTx at -4 ºC in pH 7,4 with 500mM NaCl, 1M (NH4)2SO4, 60mM of threalose and 10% glycerol, and at -80 and -196 ºC with glycerol 10% were effective in maintaining the hemolytic activity. Furthermore, the hemolytic activity is calcium dependent and completely inhibited by EDTA. Cholesterol and phosphatidylglycerol reduced the hemolytic activity of Sp-CTx which suggests that it is a barrel-pore-forming toxin and that the pore formation is dependent of electrostatic interactions with membrane lipids. Through western blot, using the “DIG Glycan Differentiation” kit, sugar motifs of Sp-CTx were evaluated, which suggested the presence of: galactose (1-4) N-acetylglucosamine and sialic-acid (2-3) galactose in N- and/or O- glycan complexes. Using the mouse hindpaw test it was shown that Sp-CTx induced intensive nociception and dose-dependent edematogenic responses in the mice footpad Sp-CTx. The local induced edema was considerably reduced by previous administration of aprotinin and HOE-140. These data suggest the involvement of the kinin– kallikrein system in the inflammatory response induced by Sp-CTx.

New isoform contain in addition of loss of N- terminus hexapeptide (as found in parent molecule β-trypsin) an intra-chain split between Lys-155 and Ser-156. The novel enzyme denominate gamma-trypsin showed similar properties with α-trypsin isoform in: polypeptide number chain (two chain), molecular masses (23,312 Da), secondary structure, hydrodynamic radius and others. In spite of enzymatic and structural similarities of both isoforms, -trypsin preferably has a lower rate formation from β-trypsin, a lower surface charge, but the gamma-trypsin has a higher thermal stability than α-trypsin. Due to obtaining facility of purification of bovine trypsin isoforms from commercial font, and properties described above, become this enzyme an interesting alternative for the food industry, detergent and biocatalysis research.

Composed group under analysis, 40 patients attended the Basic Health Unit of Consolation, Vitória / ES, aged 25 to 80 years, diagnosed with DM2 and that were already evoluted to insulinization. As controls, 40 patients were selected in the same age group without laboratory and / or clinical diagnosis of DM. Markers of inflammation, hypercoagulability and fibrinolysis were measured: Fibrinogen, D-dimer (D-Di) and plasminogen activator type 1 inhibitor (PAI-1). The polymorphism (-675 4G/5G) in the promoter region of the PAI-1 gene was correlated with their serum levels. Biochemical parameters were measured: plasma glucose (PG), glycated hemoglobin (A1C), total cholesterol (TC), HDL cholesterol (HDL-C), LDL (LDLc) cholesterol, triglycerides (TGC), ultrasensitive C reactive protein (hsCRP), urea and serum creatinine. There was still checking BMI, obesity, smoking, hypothyroidism, hypertension, dyslipidemia and insulin resistance. Statistical analysis showed significant differences (p <0.05) between groups with regard to mean values of HDLC, VLDL-C, TGC, urea, hsCRP and fibrinogen. There was significant difference between groups for VLDLc, TGC, creatinine and fibrinogen. Controls present correlations between: fibrinogen and glucose, hsCRP and fibrinogen, PAI-1 and glucose, PAI-1 and BMI. In insulinization DM2 group correlation was observed correlation between: fibrinogen and D-di, hsCRP and fibrinogen, D-Di and glucose (negative), PAI-1 and triglycerides, PAI-1 and BMI. PAI-1 levels were higher in the control group in subjects with genotype 5G5G, 4G5G and 4G4G followed. Binary Logistic Regression confirmed that the variables hypertension and fibrinogen were significant at p-value (0.009) and (0.049) and adjusted odds ratio (4.184, 1.426 to 12.276) and (3.293, 1.006 to 10.775), respectively, showing that hypertensive patients have a risk 4.18 times more likely to have insulinization type 2 diabetes and that individuals with hyperfibrinogenemia have a 3.29 times greater risk. With this study we hope to contribute to better understanding of the complex changes that accompany the user insulinization type 2 diabetic patients in expectation of seeking appropriate treatment and prevention for the macrovascular complications of diabetes.
