Efeito do acetato de medroxiprogesterona sobre o desenvolvimento das glândulas endometriais em cadelas: avaliações histológica e imunoistoquímica.

Bitches with uterus devoid of endometrial glands, phenotype called uterine glandknockout (UGKO), should be sterile animals, and could contribute to the species overpopulation control. Since the inadequate exposure of the female reproductive system to steroids during critical periods of development can lead to the formation of this phenotype, as observed in cows, sheep and rats, the aim of this study was to evaluate the effect of serial applications (from birth to 6 months old) of medroxyprogesterone acetate on the development of endometrial glands in the bitch. Sixteen cross-bred females, from different litters, were distributed in the MPA group (n = 8), females that received injections of medroxyprogesterone acetate (10mg/kg/SC), every 3 weeks, from day 1 after birth to age of 6 months, and C (n = 8), control females, untreated, that received injections of 0.9% NaCl solution, treated similarly. At age of 6 months, ovariohysterectomy was performed in all animals, and samples of uterine horns were collected and fixed for histologic and immunohistochemical analyzes. We measured the total thickness (µm) from the uterine wall, endometrium (µm); myometrium (µm), diameter of the endometrial glands (µm), number of endometrial glands / mm2, epithelial height (µm) and expression of ER –a, ER-ß and PR in the surface epithelium and endometrial stroma. Bitches from MPA group showed 35% reduction in the number of endometrial glands, endometrial glands with larger diameter, greater uterine wall, endometrium and myometrium thickness, and the height of the epithelium. There were no statistical differences between groups in relation to the expression of ER-a, ER-ß and PR in both the surface epithelium and in endometrial stroma. It is concluded that serial applications of medroxyprogesterone acetate, from birth to 6 months old are not able to cause total ablation in the development of endometrial glands in the bitch, but a 35% reduction in its development.
